Macrophage Response Driven by Extracellular ATP

چکیده

The purine nucleotide ATP is a fundamental unit in cellular energy metabolism. Extracellular and its metabolites are also ligands for family of receptors, known as purinergic which expressed ubiquitously almost every cell type. In the immune system, extracellular signals regulate migration activation cells to orchestrate induction resolution inflammation. this review, we provide an overview receptors their downstream signaling related macrophage activation. We discuss roles functions physiological pathological conditions.

Extracellular ATP protects against sepsis through macrophage P2X7 purinergic receptors by enhancing intracellular bacterial killing.

Extracellular ATP binds to and signals through P2X7 receptors (P2X7Rs) to modulate immune function in both inflammasome-dependent and -independent manners. In this study, P2X7(-/-) mice, the pharmacological agonists ATP-magnesium salt (Mg-ATP; 100 mg/kg, EC50 ≈ 1.32 mM) and benzoylbenzoyl-ATP (Bz-ATP; 10 mg/kg, EC50 ≈ 285 μM), and antagonist oxidized ATP (oxi-ATP; 40 mg/kg, IC50 ≈ 100 μM) were ...

ATP synthesis driven by proton transport in F1F0-ATP synthase.

Topical questions in ATP synthase research are: (1) how do protons cause subunit rotation and how does rotation generate ATP synthesis from ADP+Pi? (2) How does hydrolysis of ATP generate subunit rotation and how does rotation bring about uphill transport of protons? The finding that ATP synthase is not just an enzyme but rather a unique nanomotor is attracting a diverse group of researchers ke...

Paracrine stimulation of surfactant secretion by extracellular ATP in response to mechanical deformation.

We developed a heterologous system to study the effect of mechanical deformation on alveolar epithelial cells. First, isolated primary rat alveolar type II (ATII) cells were plated onto silastic substrata coated with fibronectin and maintained in culture under conditions where they become alveolar type I-like (ATI) cells. This was followed by a second set of ATII cells labeled with the nontrans...

Nonlinear fluorescence response driven by ATP-induced self-assembly of guanidinium-tethered tetraphenylethene.

Nonlinear fluorescence response, which is particularly important to attain the high signal-to-background ratio, was realized by the aggregation-induced fluorescence increase of guanidinium-tethered tetraphenylethene with ATP.

Assessment of extracellular ATP concentrations.

Most cells release ATP to the extracellular milieu. Extracellular ATP plays important signaling roles by activating a score of broadly distributed cell surface purinergic receptors (purinoceptors). Biological responses regulated by purinergic receptors include neurotransmission, smooth muscle relaxation and contraction, epithelial cell ion transport, inflammation, platelet activation, immune re...
